Question

Cocaine is extracted from a dollar using two different techniques. Technique 1 used only methanol for extraction. The dollar bill was submerged into methanol and shook. The Solution was then rotovaped. It was redissolved in methanol. Technique 2 used HCl NH3 MEOH, and dichloromethane to extract Cocaine. The dollar bill was submerged in HCl. NH3 and dichloromethane was then added to solution and shook. The organic layer was then rotovaped and redissolved in MEOH. Is one extraction technique expected to be better than the other? Why? (Consider which formof coacaine is more appropriate for GCMS.

Explanation / Answer

Cocaine was extracted from a dollar using methanol extract by solid-phase extraction. The solid phase extract was analyzed by gas chromatography mass spectrometry in full scan mode and quantitation was acheived in selected ion monitoring mode deuterated internal standard.

Although cocaine and cocaethylene can be analyzed by GCMS without derivative, benzoylecgonine must be derivatized prior to analysed by GCMS.Benzoylecgonine is a hydrolysis product of cocaine.
