Crystal voilet and safranin are the dyes used to differentiate gram positive and gram negetive bacteria. Gram negetive bacteria unable to retain crystal voilet due to a very thin layer of peptidoglycan or peptidoglycan is absent. But human cells don't posses peptidoglycan and it is useless to use gram stain on human cells. But yet if we want to perform then crystal voilet is not retained and cells should stained with safranin to give pink colour.
