In both the cases, Glycophorin A is only partially digested by trypsin.

ROV will have the most portions of glycophorin outside, due to the SDS gel protein band. ROV+(small protein size) has the lowest band, which implies that only the large part of the protein was digested by trypsin. IOV+ (big protein size) has a higher band implying that only a small portion of the protein was digested by trypsin. As for Glycophorin A, its major portion is inside the erythrocyte and its minor portion is on the outside of the erythrocyte.
