Question

Refer to the figure below (improved version of your lab run!) for the following questions: a. Which of the lanes (left, center, or right) contains the control (undigested) b. Do you think the plasmid digested as expected for the experimental c. Using the DNA ladder key provided below and/or the restriction map on p. sample? [1] sample? [1 9, what size (approximately) is the band on the bottom in the right-most lane? For ladders, it is best to determine band sizes from the bottom up- the bright band on the bottom is 100 bp (or 0.1 kb). 12] 1000 45 900 45 900 45 600 45 9 500 115 23 400 40 8 300 40 8 - 200 40 8 100 40 8 0.5 uglane, 8 cm length gel, 1X TBE, 5 V/cm, 1 h Figure 1. Explanation / Answer

The middle one is undigested since undigested will continue to have a supercoiled form and not migrate to after a certain length or size. Howver the digested sample is open and fragmented that will give you three bands when run and visualized. Yes, the digested sample has given three bands which depicts the three forms of DNA sample when digested and visualized. The bottom band in the right-most lane is approx 100 bp according to the ladder.
