Question

If you want to reproduce the shuttling experiment as performed during week 3. you will need to purchase Cycloheximide (CHX) from one of three potential suppliers. CHX has a molar mass of 281.35 g/mol and can be dissolved in DMSO, water, or EtOH but with different solubilities. You will need to use CHX at a final working concentration of 36 g/mL in a 100 mm dish with a total media volume of 10 mL. Determine how much of the stock solution purchased from each company you would need (inHL) to add to the media in the 100 mm dish to obtain the final working concentration of CHX. Give your answers in L using the scientific notation. a. Company A supplies CHX dissolved in DMSO at 100 mg/ mL: Volume needed in L: 36 Submit Answer Answer Submitted: Your final submission will be graded after the due date. Tries 1/99 Previous Tries b. Company A supplies CHX dissolved in water in 2% solution volume needed in L: 1.8 Submit Answer Answer Submitted: Your final submission will be graded after the due date. Tries 1/99 Previous Tries C. Company A supplies CHX dissolved in EtoH at 50 mM Volume needed in L: 2.559 Submit Answer Answer Submitted: Your final submission will be graded after the due date. Tries 1/99 Previous Tries

Explanation / Answer

Working concentraiton for CHX is 36µg/ml

we need total volume of 10ml

so in 10 ml the total concentraiton of CHX is 360µg

A)now company A has CHX at DMSO at 100mg/ml

or 100 X 1000µg present in 1ml or 1000µl

then 360µg will be present in 1000 X 360/ 1000 X 100= 3.6µl

B)

2% means in 100ml we will have 2gm of CHX

Concentration is 2gm/ 100ml 2X 1000mg / 100ml or 20mg/ml

Now we require 360µg

Then 20X 1000µg is present in 1000µl

360µg will be present on 360 X1000 / 20X 1000 =18µl

c) CHX concentration is 50mM

Molecular weight of CHX is 281.35g/mol

Thus to make 1000ml 1M is 281.35gm

                         10ml 1M .. 281.35 X 10/ 1000= 2.81 gm or 2.81 X 1000mg or 2810X 1000µg

2810X 1000µg----- 1M

Then 360µg--------1M X 360/ 2810X 1000 or 1000X 360/ 2810X 1000mM= 0.128mM or 128µM

Now stock concentration is 50mM or 50X 1000µM

Dilution 50 X 1000/ 128 = 390 fold

Thus for 10ml we require 10 X 1000 / 390 = 25.6µl
