In the manuscript by Dolatshadet al. manuscript that we discussed in class, which described two families of GPCRs (S1P_1-5 and LPA_1-5) used to maintain and promote survival of human embryonic stem cells. This review states that loss of signaling through Gi alpha led to a change in the way that embryonic stem cells interact. Specifically, it changed their interactions. Describe how the signaling through Gi alpha was disrupted to demonstrate the role for the protein in maintaining stem cells. It is seen based on researches that activation of the Gs-cAMP signaling pathway in mESCs contributes to the maintenance of transcription factor expression which is important for pluripotency ( Layden, 2010)). In addition, evidence exists for the involvement of the cAMP/PKA pathway in mESC self-renewal pathways ( Faherty, 2007). Further, Gi- and ERK-dependent mechanisms leading to the activation of pro-survival pathways, apoptosis inhibition and increased proliferation (Pebay). Hence any mutation leading to a nonfunctional Gi leads to down regulation of these pathways and hence can lead to loss of stemness.
