In this assignment you have to design a gene expression microarray experiment with 50 spots on the chip. First you can choose the study question from your recent project or a question interest you. I have some potential questions as suggestions below also just in case you are not sure about a good question. First introduce the problem with a short introduction (min 250 words). You have to include references here after every statement. There will be 2-point deduction after each missing references or reference with a wrong format, (see below format requirements), if you do not have any reference in your introduction it will results 10-point deduction. If you have your introduction less than 250 words it will be one point deduction /10 missing words. Next select specific genes (at least 10), which should have already proven in the literature to have function in the mechanisms of the gene regulatory pathway you study (if you study estrogen responsiveness it should be genes which are expressed when you expose your cells to estrogen). Please refer Module 6A discussion on Alzheimer disease for ideas. The selection of each gene has to be explained by a min 50-word statement plus with an appropriate peer-reviewed primary reference (NOT A REVIEW PAPER) from PUBMED or PUBMED CENTRAL. Non-peer-reviewed reference or a URL will not be considered as an appropriate reference. The use of any non-peer-reviewed reference or URL (website) results in 2 points deductions/reference. Not providing name, description or function also results 2 points/gene. You also have to choose control genes for this microarray chip. Again, be sure you understand what kind of control genes you need for this chip, how many and where should the located in the chip (please refer to my microarray lecture I provide a lot of explanation what you need to do for control genes. You have to decide how many spots you will dedicate for the controls (positive and negative). Please be sure your control gene are not just housekeeping genes (remember negative controls!) At the end of the assignment, you need to include a chart, which indicates the position of each gene (provide position such as: A1 human actin, A2 human estrogen receptor …etc). Be sure your control probes are at the right positions (refer to our microarray lecture). Missing chart will result a 5-point deduction.
