Question

You are involved in a research team working on how mutation affects the function of a genome. The research group is now focused on how the mutations on the upstream of the open reading frame (ORF) affect the expression of a specific gene.

1. Generate a hypothesis on the potential effect of mutations on how the mutation on the upstream of a gene can alter the gene expression.

2. Design experimental strategy of mutational analysis and describe on any anticipated result (predicted outcome from your experimental design).

Explanation / Answer

1). The upstream of the gene (towards the 5’ end of the sense strand) contains the promoter regions prior to the transcription starts. A mutation in the promoter region of an operon would most likely cause the promoter to prevent binding to RNA polymerase. This results in inhibition of gene expression.
