Question
Can someone help me with questions 1 and 2
1. Perform the necessary calculations for part I of the protocol. (Hint: weight/volume concentrations tell you the weight of solute per 100 ml of solution. For example 200 ml of a 5% (w/v) NaCl solution would be made by dissolving 10 grams of NaCl in 200 ml of water.) 2. Draw what the following PCR samples would look like when run on an agarose gel. Each sample (a-f) should be in it's own lane/column on the gel. a) Non-GMO plant with Plant master mix b) Non-GMO plant with GMO master mix c) GMO plant (with both CaMV promoter and NOS terminator) with Plant master mix d) GMO plant (with both CaMV promoter and NOS terminator) with GMO master mix e) GMO plant (with CaMV promoter only) with Plant master mix f) GMO plant (with CaMV promoter only) with GMO master mixExplanation / Answer
Ans- 1.5g of Agrose+ 5ml TAE+ 45ml of DI water
Calculations- 3% agarose means 3g of agarose dissolved in 100ml of DI water hence to make 50ml of gel 1.5g agarose will be dissolved in 50ml TAE
Now to make 1X TAE from 10X TAE we have to dilute the stock i.e 10X TAE 10times. Hence 1ml of TAE is mixed with 9ml of DI water to make 1X TAE. So to make 50ml of 1X TAE 5ml of 10XTAE+ 45ml of DI water.
