15. Thioglycollate broth is designed to test the aerotolerance of bacteria.

It's constituents include nutrients for bacterial growth,

thioglycollic acid, thioglycollate, L- cysteine to reduce oxygen to water

And methylene bland am indicator for presence of oxygen

and agar which helps in retarding diffusion of oxygenn and helps in maintaining the different layers of broth containing different organisms.

This is the reason why this medium is used to measure aerotolerance.

Through autoclave get, oxygen diffuse out of the medium but when the medium sits at room temperature oxygen starts to diffuse in again that is why it is important to use this medium soon after autoclaving.
