Western blotting is a technique for the separation of protein through gel electrophoresis. In western blotting a probe is therefore an antibody that binds specific target protein.

Difference between western blot and immunoflourescence :

Western blotting is a technique also called as immuno protein blot. It is used for the separation and identification of specific protein from a cell or tissue extract. Here, proteins are separated through gel electrophoresis and then hybridized with antibody (probe).

On the other hand, immunoflourescence is a technique which is used for microscopic observation of protein, their location and distribution within the cells. In this technique a fluorescent antibody is used that reacts with the epitope of antigen within the cells and flouresces when observed under fluorescent microscope.
