I have purified Delta and have an assay to determine if it binds Notch. I know after my purification steps Delta is in its native shape and binds Notch as my binding-assay gives a positive response In a separate assay, I add beta mercaptoethanol to my Delta before binding to Notch. I ne longer get a positive response from my binding assay What is likely to have happened? O I have unfolded and disrupted all secondary and tertiary protein structure have desturbed tertiary structure by disrupting disulfide bridges I am breaking peptide bonds cutting the protein into pieces have interfered with hydrogen bonding in the backbone and disrupted secondary structure and thus tertiary but left covalent bonds intact I haven't done any of these things
