Explanation / Answer

1.Since the photograph is not provided the first part of the first questions cannot be answered.However, if the colonies are obtained as single colonies far from each other as isolated colonies the isolation was successful. If not, The isolation may have not been successful due to the following reasons, the culture being to dense hence matt growth may have appeared .hence dilution of the sample may have too be done.

2.The new culture will be considered pure only if a single colony was picked up and inoculated. If a loopful from the matt growth is picked it is not considered a pure culture. A single colony is formed which represents one single organism hence only that can be considered a pure culture.

3.A pure culture could become a mixed culture by cross contamination . improper sterilization of loop mixing of meda etc. It could be problematic if DNA extraction or chemical test are to be done on the culture the results could not be dependant on.
