Moreover, you can choose the concentration of ADP you want to use in your experiment [zero (none), 500 nmol, 750 nmol or 1000 nmol]. Furthermore, you can chose among five different metabolic inhibitors of cellular respiration (amytal, antimycin, cyanide, oligomycin, or rotenone) to add to your experiment. Lastly, you can decide to add the uncoupler DNP [2,4-dinitrophenol].

The electrode will measure oxygen concentration in the experiment. Oxygen consumption in the experiment will be determined as a measurement of the progress of reactions against time.

Hypotheses for Experiment #1:

1) Do you predict that oxygen consumption will increase, decrease or not change at all in the reaction flask after the addition of succinate alone? Why? Recall that succinate is a substrate in the citric acid cycle.
