Explanation / Answer

A. Western blotting data.

Comparison has been done between control and K5-R1/R2 mice at the age of P18 and 5 months. The activation of proteins has been looked at by the levels of phophorylated forms of Stat3, p38, NF kappa B. Phorphorylation of NF kappa B increases in mice at 5 month as compared to the controls. In p18, the activation od NF kappa B does not occur. The Phospho forms are normalised. TO the non Phospho forms of the proteins. Similarly, stat3 Phospho levels increase when mice are at 5 months age. And also, tje levels of p38 increase in P18 and 5 months old mice in K5-R1/R2 when compared with the controls.

B. Real time data has been shown which deals with the abundane of transcripts. Inflammation markers have been been taken. The abovementioned cytokines increase in K5-R1/R2 mice as compared to the controls. In both 5M epidermis and 8M dermis, expression of inflammatory cytokines increase by several folds compared to the controls.
