Question

IfT-39%, what is the percent A. C, C? If A=19%, what is the percent T, C, G? What kind of bond holds the backbone of a single strand of DNA together? R. Orr, 2018 What kind of bond holds the DNA double helix together? What is meant by the term “antiparallel?" Practice building a daughter strand of DNA using this GAME. Review the concept of DNA by clicking on this TUTORIAL and then click on the "DNA" link listed on the page Outcome 2 Watch DNA Replication (schematic) Practice building your own DNA using this INTERACTIVE simulation. What determines the sequence of the daughter DNA? What is meant by the term "semi-conservative" replication?

Explanation / Answer

Ans. Chargaff’s rule for dsDNA-

No. of A nucleotides = No. of T nucleotides,

Or, A (purine) = T (pyrimidine) -----equation 1

No. of G nucleotides = No. of C nucleotides,

Or, G (purine) = C (pyrimidine) ------ equation 2

#1. Given, T = 39%

So, A = T = 39%

Now, G + C = 100% - 39% = 61%

Using G = C   -

            G + G = 2G = 61%

            Hence, G = 30.5 %

Therefore, G = C = 30.5 %

#2. Given, T = 19%

So, A = T = 19%

Now, G + C = 100% - 19% = 81%

Using G = C   -

            G +G = 2G = 81%

            Hence, G = 40.5 %

Therefore, G = C = 40.5 %

#3. The backbone of single strand of DNA is formed by phosphodiester bond between adjacent nucleotides.

#4. Hydrogen bonds between complementary nucleotides on opposite strands keep the two strands of double helix together.

#5. One strand runs in 5’-3’ direction whereas the other complementary strand runs in 3’-5’ directions. Since the polarity of two strands is opposite- i.e. 5’-end of one strand faces 3’-end of the complementary strand, the two DNA strands in s dsDNA molecules are called antiparallel to each other.

#7. The sequence of parental DNA determines the sequence of daughter DNA. The sequence of daughter DNA is complementary to that of parental DNA strand.

#6. In semiconservative replication, each daughter dsDNA inherits one strand from its parental DNA and the other strand is newly synthesized. That is, one strand out of total 2 strands in the new dsDNA molecule is conserved (inherited from parental DNA).
