Describe in sentences or in bullet-points the main components (coordination number, geometry, metal ion, oxidation state, types of ligands, etc) of the active site for DNA polymerase
and
Include a picture of another form of the active site (oxidized or reduced or other) of your enzyme. Describe the main features and include at least one reference.
NH2 NH2 N dCTP substrate primerN terminus cleavage of pyrophosphate group 3' nucleophilic attack OH (postulated) H2o... ..?Mg. 2Mg Asp192 H20 Asp256 Asp190Explanation / Answer
The main components of the active site of DNA polymerase include three acidic residues that are conserved which would be binding two the DNA polymerases. The core of the DNA polymerase contains a motif A, in which the amino acid residues present at positions 605 to 617 are important. This motif A residues interact with the dNTPs that are coming in. The Asp 610 would be reacting with the Mg2+–dNTP complex. There is a motif B. This is present from positions 656 to 670.
3’ to 5’ exonuclease activity has conserved domains Asp 249, ser 252, leu253, thr 254 as well as pro 255.
