Question

Please answer questions 1 c) and 1 d) thank you. Here is additional information to help solve 1 d)

12.2G Synthesis question: Wild-type mice are adoptively transferred with naive CD4 T cells isolated from OT-II T cell receptor transgenic mice. These OT-ll T cels are specific for a peptide of the chicken ovalbunin protein (OVA) bound to MHC class II. After transfer, he OT-ll cells rapidly disperse to all of the secondary lymphoid organs in the recipient mice. Half of the recipient mice are then given OVA protein dissolved in their drinking water, and the other half of the mice receive normal drinking water for 5 days. On the following day (day 6), ymphoctes from the spleen, the peripheral lymph nodes (LN) mesenteria lymph nodes (MLN), Peyer's patches (PP) and lamina propia (LP) are isolated and examined for the proportions of CD4 T cells in each organ expressing FoxP3. The results are shown in Figure 012.26A Figure 012.26A a) What is the conclusion from this experiment? To investigate this further, dendritic cells are isolated from the spleen (spl DC) or from he lamina pcopcia of the small intestine (LP DC). Each subset of dendritic cells is pulsed with the OVA protein, and then incubated with naive OT-II CD4 T cells for 5 days in the presence or absence of TGF-?. The CD4 T cells are then examined for FoxP3 expression; the results are shown in Figure Q12.2GE TGF Figure Q12.26B

Explanation / Answer

1. c)

1. The major difference between LP derived dendritic cells and splenic dendritic cells is that the splenic dendritic cells are CD103+ CD11b+ whereas the LP dendritic cells and CD103+ CD11b-.

2. The devwldevelo of LP dendritic cells depends on BAT3- leucine zipper factor (ATF like), ID2- Inhibitor of DNA binding and IRF8- IFN regulatory factor. Splenic dendritic cells' development does not depend on these factors.

3. Splenic dendritic cells show high expression of SIRPa and DCIR2- dendritic cell inhibitor. LP dendritic cells do not express these factors.

1. d)

Out of the given four groups of cells, Peyer's Patches and Lamina Propria qouwo show high expression of a4b7.
