Question

Can someone please explain how to obtain the distances. Will give a thumbs up

1 An RFLP is identified in a banana slug. Digesting DNA with EcoRI and using a specific probe generates either a large 7000 bp restriction fragment or a small 2000 bp restriction fragment. The size of the slug is regulated by the gene L. Large (L) is dominant to small (1). The color of the slug is regulated by the gene Y. Yellow (Y) is dominant to brown (y) Atnue breeding large vellow slug with the 7kb RFLP is crossed to a small brown slug with the 2kb RFLP The FI heterozygous slugs are test crossed with a small brown slug with the 2kb RFLP and a thousand progeny are scored. Large Yellow 7 & 2 kb-RFLP Large yellow 2kb-RFLP Small yellow 2kb-RFLP Small brown 2kb-RFLP Large brown 2kb-RFLP Small vellow 7 & 2'kb-RFLP 362 87e 358 SmaH brown 7 & 2 kb-RFLP 115 Large brown 7 & 2 kb-RFLP. 93 Is the RFLP linked to either of these two genes? (show your calculations) If the RFLP is linked, draw a map with the RFLP and the linked gene/s (show your calculations) Large.10MU-RFLP-20MU-Yellow dag e dominant tz: progeny list ivo. ) 2011000 ??100 2.1 Ditu nie ke twee un UW 12

Explanation / Answer

According the given question the highest values are 362 and 358 respectively, these are the parental types

And the least values are due to double recombinant: 9 AND 11

The single recombinants are: 87, 43, 37 and 93 respectively

Distance between large and RFLP = total recombinants/total products

= 9+11+87+37/1000 = 144/1000 = 0.144 = 14.4 mu

distance between RFLP and yellow = 9+11+43+93/1000 = 0.156 = 15.6 mu

between yellow and brown = 9+11+37+43 /1000= 0.1 = 10 mu

SO the distance map will be

Large-------15.5-------RFLP-----14.4-------yellow
