The 2-dimentionslal double diffusion immunoprecipitation. experiment (Ouchterlony plate) did not work well this semester. This display is similar to the one attempted in lab. Each well has a hexagon shape. Please indicate which samples were placed in each well to generate the precipitation bands displayed. Center well contains: Anti-human whole serum developed in a goat

Explanation / Answer

Generally Ouchterlony double diffusison is a technique used in the detection and the identification of antigens and antibodies.

Antibodies should kept in one well and different samples kept in different wells so as to confirm about the antigens present in the samples. If the samples has antigens for that perticular antibody, they must precipitate and form a band, as shown in the figure.

In the given problem, the centre well contain anti human whole serum which react specifically with the normal human serum.

Thus, the well 4 contain human whole serum,

Since anti human whole swrum does not react with any of the immunoglobulins, thus well 1, 2 and 6 contain human IgA, IgG and Human albumin.

Well 3 and well 5 contain fetal human whole serum.
