Question

Please help answer the following question about the subunits of a protein complex within the polyacrylamide gel. Please help explain why you chose your answer. Thank you .

1. Three subunits of a protein complex have been resolved in a two-dimensional polyacrylamide gel, as shown in the schematic drawing below, Indicate whether each of the following better describes the a b, or c subunit. Low pH pH gradient. High pH Co e80 i.( ) It has the smallest apparent molecular weight ii.( ) It has the highest negative charge iii.( ) It has the lowest isoelectric point.

Explanation / Answer

(i) "b" has the smallest apparent molecular weight. Because, lowest molecular weight subunits move at faster rate (the molecular weight decreases in the direction of SDS migration).

(ii) "a" has the highest negative charge. Negatively charged subunts move towards anode (low pH conditions exist near anode).

(iii) "c" has the lowest isoelectric point. Molecules migrate along with gel until they reach isoelectric point,.
