Question
Mid-Term Exam BCH 4034 L (Biochemistry II Lab Explain (in 3-4 sentences) how ampicillin functions as an antibiotics? Explain how ampicillin resistance is accomplished by the plasmid DNA containing an ampicillin resistant gene. (5) 4. 5. You have two LB-agar plates. One contained no ampicillin and the second plate contains ampicillin. You transform bacteria with a plasmid that contains an ampicillin resistance (Amp) gene. You then streak both LB-agar plates and incubate at 37°C. Draw what you should observe the next day when you pick up the plates. Explain. (3) LB Agar LB Agar + Ampicillin 6. What is genetic engineering? Briefly explain the importance and practicality of using genetic engineering to life in general. (2)Explanation / Answer
Ampicillin mechanism
it has the ability to penetrate Gram-positive and some Gram-negative bacteria. Beta-lactam ring amino group attached to the beta carbon relative to the carbonyl group. That amino group helps the drug penetrate the outer membrane of Gram-negative bacteria.
It acts as an irreversible inhibitor of the enzyme transpeptidase, which is used by bacteria to make the cell wall. It inhibits the third and final stage of bacterial cell wall synthesis in binary fission, causing cell lysis, therefore, ampicillin is bacteriolytic.
ampicillin resistance mechanism:
beta-lactamase produced by the bacteria attaches to the beta-lactam ring of antibiotic and opens the ring this prevents attachment to transpeptidase, therefore, the bacteria can function normally forming cell wall synthesis
