Question

Two proteins (A and B) were separated by chromatography on a cation exchanger column (pKa ~4). The load flow-through, wash, and eluted fractions were collected such that both proteins were detected (via A280 readings) at some point in the experiment. The chromatograms obtained this way are shown in Figure 3.

Answer the following questions with True or False. If a statement is false, explain WHY. No credit will be given is false statements are not explained.

a) Protein A is either negatively charged or neutral. (8 pts.)

b) Protein B is negatively charged. (8 pts.)

c) The interaction between protein A and the cation exchanger was disrupted by 0.2 M of ammonium sulfate. (8 pts.)

d) Protein B was eluted from the column at 0.3 M ammonium sulfate. (8 pts.)
