Question

The N-terminal tryptic peptides from the beta chains of HbA and HbS are as follows: HbA: Val-His-Leu-Thr-Pro-Glu-Glu-Lys HbS: Val-His-Leu-Thr-Pro-Val-Glu-Lys
a. Would these peptides separate from each other in an electric field at pH 7?
b. What is the approximate net charge on each peptide at pH 7? The N-terminal tryptic peptides from the beta chains of HbA and HbS are as follows: HbA: Val-His-Leu-Thr-Pro-Glu-Glu-Lys HbS: Val-His-Leu-Thr-Pro-Val-Glu-Lys

b. What is the approximate net charge on each peptide at pH 7?

Explanation / Answer

a) yes we can separate both at pH - 7

difference between both peptides are Glu in HbA Val in HbS

Glu is acidic amino acid while Val is neutral Glu having negetive charge at pH 7 so it can move but Val will not movie either side. so we can separate.

b) in HbA one negetive charge will be there. HbS no net charge .
