Question

Complete the following dilution and titration problems. Show calculations a) A 3-fold serial dilution is performed 5 times. What is the final dilution and correction factor for the last tube? b) Determine the concentration of an 800mg/dL sample after the following dilutions have been performed: (round to the nearest hundredth) 1/4,1/12,1/8,1/4 c) Determine two ways to make the following final dilution: 1/180 d) Determine a patient's antibody titer if the first 4 tubes of a 6-fold serial dilution show a reaction.

Explanation / Answer

ANSWER:

a) In serial dilution the final dilution is obtined as

Dt = D1 X D2 X .... Dn

Dt= final dilution Di's = dilution factor

here dilution factor is three and this dilution is performed 5 times therefore

Dt = 3 X 3X 3X 3X3 = 243

so final solution will be 1/243 dilute

b) Use the following equation todetermine final concentration

Cfinal = Cinitial/D   D = dilution ratio = D2 X D2 XD3 X.....

D = 4X12X8X4 = 1536

Cfinal = 800/1536 = 0.521mg/dL

c) 1/180 dilution can be obtained as

i) make 1/10 first then 1/9 and finally 1/2

Dilution ratio = 10X9X2 = 180

ii) Dilute as 1/2, 1/5, 1/9, 1/2

total dilution = 2X5X9X2 = 180
