Question
For the following enzyme Lysozyme:
1) for the reaction catalyzed by the enzyme: what are the reactants? what are the products?
2) define the reaction as uni-uni, uni-bi, bi-uni, or bi-bi. Briefly explain
3) Does the enzyme follow a sequential or ping-pong reaction coordinate? Briefly explain
4) Describe the catalytic mechanism of action for lysozyme. include specifics about the amino acid residues involved & how they contribute to catalysis. Define any "hallmark" catalytic mecahism(s) used by the enzyme.
Explanation / Answer
1: Lysozyme is hydrolase enzyme that stimulates the hydrolysis of the bond between N-acetylglucosamine(NAG) and N-acetylmuramic acid(NAM) present in the polymer peptidoglycans.
Hence the reactant for lysozyme is peptidoglycans(a polymer of NAM and NAG) present in bacterial cell wall and the product is a disaccharide derivative which is NAM - NAG
2: Since the substrate is one (peptidoglycan) and the product is one (disaccharide derivative NAM - NAG), this is an example of uni - uni reaction.
3:The reaction mechanism follows a ping-pong reaction co-ordinate as chemically modified form of the enzyme hydrolase are formed as intermediate and take part during the reaction.
