Hey everyone,I am currently undertaking independent research in which I grow a strain of bacteria in different types of agre to calculate cell growth using fractional analysis. So this includes a box count method obviously.
This lab really includes both knowledge of Microbio and Physics. But I had a problem, and I was hoping somebody here with some vast knowledge could lead me in the right step of taking a CLEAR picture.
I am using a program called "FracLac" for use of box counting http://rsbweb.nih.gov/ij/plugins/fraclac/FLHelp/UseFracLac.htm
I need to get a clearer picture of our bacteria so that we can achieve more accurate of a box count for our calculations. We have tried using a microscope and taking pictures, and even using a scanner (as done in similar research in the 90's by Japanese scientists). However, the quality is not what I am looking for.
Does anybody know a method in which I can take a very good quality picture of the bacteria? I would appreciate anything that is feasible. If you offer me an idea that would be impossible to achieve though, like getting an electron microscope or something, please let me know. I accept all ideas.

^ that is the kind of HIGH quality I want. I REALLY want defined pictures, as little fuzzy as possible.
