Please answer all the three questions. Thanks.
10.16 How does a cell's transcriptome compare with its proteome? a. For a specific eukaryotic cell, can you predict which has more total members? Can you predict which has more unique members? Suppose you are interested in characterizing changes in the pattern of gene expression in the mouse nervous system during development. Describe how you would efficiently assess changes in the transcriptome from the time the nervous system forms during embryogenesis to its maturation in the adult. How would your analyses differ if you were studying the proteome? b. C.Explanation / Answer
Answer10.16:
Transcriptome is all RNA content of a cell at a given time . it is dynamic in nature that is it varies with cell to cell, tissue to tissue and organ to organ with time. but proteome is very much similar to transcriptome but here we talk about gene expression at protein level.
Transcriptomics is studied by using DNA microarrays and proteome is studied by electrophoresis and mass spectrometry.
Anawer a) Among the eukaryotes to to complaxity in structure and persence of interons as well as exons the multicellularity has evolved at least 25 times then to unicellular ancestors.Complex multicellularity has evolved ten times in eukaryotes , once in Animalia, three times in Fungi , and six times in the three major photosynthetic clade .
Answer b) For analyzing the developmental pattern or gene expression level for any of developmental stage we should follow the following pattern....
1. to produce DNA chip
2.take and measure the transcriptome or total mRNA content of cell.
for step 1 we must design the chip of the same organism which we are using for getting the mRNA.Now take the DNA ----- amplify the DNA using PCR----- denature the DNA------ SSDNA------- put this SSDNA in grooves od DNA chip.
for step2.we have to extract mRNA of cell at perticular stage whose expression we want to check.
cell---mRNA------cDNA through reverse transcriptase----- now we have cDNA which is also SSDNA.
now we use hybridization method and also use some dye to examine the expression of gene .for this we generally use Cy3 and Cy5 dye.after whole setup we can examine it to alalyzer.
Answer.3 When we are dealing with proteome then protein expression is analyzed by using electrophoresis and mass spectrometry.
