Question

Describe your observations for the concentration of extracellular glucose in growth medium from the table below for both strains grown under the three different conditions:

Concentration of Extracellular Glucose in growth medium Growth time of cultures Cell concentration cells/mL rowth (Tm 3.75 mM No. Carbon O2 Strain of EtOH Presence at cessation of Initial (To)Final (Tm) source 30 1Glucose 8.450 0.022 0 20 Yes 0.35 8.399 0.017 20 2 Glucose Yes 0.55 8.637 0.0189 20 Yes 0.5 8.688 0.0183 30 Pyruvate 5.2 NIA NIA 0 NIA NIA NIA 0

Explanation / Answer

1.Glucose -present , Oxygen -present

a)In strain Gr, when no ethanol is present the final concentration of extracellular glucose is very less as compared to initial concentration. This is because glucose + water gets converted to carbon dioxide and water.Therfore, all of the glucose gets used up.

-b)In strain Pe, when ethanol is present, then glucose undergoes alcoholic fermentation and forms more ethanol and gets used up. Thats why the final concentration here is also very less as compared to intial concentration of extracellular glucose.

2.Glucose - present , oxygen -absent

In case of both strains Gr and Pe, ethanol is present , so it will undergo alcoholic fermentation as same as case 1b. As a result , here also the final concentration is less than initial one. The concentration differs by marginal numbers.

3. Pyruvate- presen, oxygen -present , Ethanol -absent

In this case as pyruvate is present as carbon source and not glucose, the cell will undergo aerobic respiration,so therefore, the extracellular glucose cannot be determined and therefore the values are not available for both the strains.
