1. The major differences between killed/inactivated virus vaccines and live attenuated virus vaccine are:
i) Live attenuated vaccines are prepared by attenuating pathogenic viruses by growing them in unfavourable conditions which result in gene mutations due to which virus looses pathogenicity but retains capacity for transient growth, whereas killed/inactivated vaccines are prepared by inactivation of the viruses by heat or chemical treatment (by formaldehyde or an alkylating agent) so that the virus is unable to multiply in the host.
ii) Live attenuated vaccines show prolonged immunogenicity and eliminate the need for repeated boosters, whereas killed/inactivated vaccines are less immunogenic so often require several boosters.
iii) Live attenuated vaccines may revert back to virulent form, whereas inactivated vaccines cannot revert back to virulent form.
iv) Live attenuated vaccines induce both humoral and cell mediated immunity, whereas inactivated vaccines usually induce humoral immunity.
