Question

Hi I did a lab on PTC genotyping using electrophoresis, gene sequencing and PTC paper. I need to write 2 paragraphs for my discussion but I need help putting it into words, below I have the instructions and my data. I was a homozygous non taster. How would I use this data to answer the questions in the instructions? Thanks

The discussion should be two paragraphs in length. The first paragraph should include final conclusions about your TAS2R38 alleles based on evidence from tasting, the gel, and sequencing. Also interpret your own genotype/phenotype; how are they related (e.g., how could a non-functional product come about) in this paragraph. Lastly, compare your genotyping and phenotyping results. Provide a biological explanation that rationalizes your genotype and phenotype results The second paragraph should report the importance and the implications of your results. What's the take home message? Are there any limitations of the study?

Explanation / Answer

The above given gene follows mendelian recessive pattern of autosomal trait inheritance. The frequency of taster and non taster allele is found vary in different populations.PTC is phenythiocarbamine responsible for bitter taste between tasters and non tasters.Test for SNP-single nucleotide polymorphic gene is responsible where ligand binds to the receptor sets an action potential that sends an impulse to sensory cortex of the brain where it is responsible for bitter taste. SNP gene has high polymorph rate of 145 length in base pairs of nucleotide length. The gene TAS2R38 allele is responsible for taste receptor. This follows Hardy Weinberg equilibrium where fraction of gene copies that are of between taster and non taster allele is found varied in the given data. The gene is targed ,amplifier, added with restriction enzymes Hae3 proceeded to incubation, which produced a heterozygous effect.

The phenotype such as homozygous is said to be normal whereas Tt which is heterozygous is said to leakist phenotype as reduced or absent.
