the product from 1 & 4confirms the presence of the insert within the lac gene. 1& 3 both are forward primers . 2 primers priming the same orientation will never amplify the gene. But 1&3 has shown a product amplification. this indicates a change in orientation of the gene.

1&2 will give a product when the cat gene is inserted in the same orientation as that of lacZ, and the 2&3 combination will give product irrespective of the orientation of insertion. But 1&3, giving product means , the orientation is changed. therefore, there are multiple copies in both orientations are inserted.
