1) The Yeast genome sequencing project finished in 1996. The complete genome contains 1.2 x 107 base

pairs.

a) If 3.6 X 107 base pairs of sequence were generated as part of the project, what fold coverage was

obtained?   1.2E7 * 3 = 3.6E7. Thus we have 3X coverage

b) With this fold coverage, what is the probability that any particular base was not sequenced?

e^-3 = 0.0497 --> 0.050% no sequenced?

1. a) The fold coverage is calculated as: C = LN/G, where C = fold coverage, L = read length, G = haploid genome length, and N = number of reads.

Fold coverage of this given genome = 3.6 x 107 / 1.2 x 107 = 3.

b) P0 = e-c = e-3 = 0.05, i.e., 5% is not sequenced, 95% is sequenced.

2. c = 1, therefore, P0 =e-1 = 0.37. 37% is not sequenced, only 63% is sequenced.
