Characteriztion of lysozyme activity- Biochemistry Lab.
a lab was performed in order to characterize isolated fractions. We were looking to isolate lysozyme. After cation resin exhange and gel electrophoresis, we used the same fractions and added them into bacterial suspension. then measured the absorbance every 30 seconds for 1 minute.
1.if there was activity does that prove that the activity is lysozyme? If not, how would you prove it? The book contains at least 4 methods I can think of right away. If you did not see activity, suggest several reasons why not.
2.How does lysozyme activity differ from the homogenate versus the high salt fractions? Include in your answer a discussion about Units of enzymatic activity/µg of protein.

2) The homogenate enzymes is the pure form of enzymes which can be obtained wheares the salt fraction is the type of purification step.. It is being found that while purification step some of the protein is lost.. The specific activity is reduced during each step thus affect the Vmax..!!
1) the lyzosome is a enzymes which is a antibacterial enzymes..!! It reduce the growth of bacteria. So, if we add the enzymes and observing it will reduce the amount of bacterial cells in the original sample thus saying that action is there.
Now,to proof it is shown by this we can preapre the same test Exctly same things except the addition of lysozyme in the bacterial culture and can observe the bacterial cell no. If it keeps on increasing with time in without lyzosome addition wheares decrease the number of cell with time in lyzosome addition. This proves that the action is shown by lyzosome
