Question

A question about microbiology. 29. (6 points): You are a lead food scientist in a yogurt company and responsible for developing a new Streptococcus thermophilus strain that is resistant to two types of phages, A and B. Currently, you have S. thermophilus A strain that is resistant to phage A only AND S thermophilus B strain that is resistant to phage B only. Using your knowledge of CRISPR system, design your method to create a S. thermophiles strain that is resistant to both phages? You are free to either modify the existing A or B strains or introduce/engineer a new S. thermophilus strain. No need to write technical details such as cloning or transformation. I just want general description in 4-7 sentences. You can use a diagram as part of your answer

Explanation / Answer

Answer : clustered regularly interspaced short palindromic repeats (CRISPR) and it's genes are responsible for acquired resistance against phages. These resistance genes has to be inserted in hypervariable regions in the bacterial genome. So the genome of S. Thermophilus has to be studied properly and in two region two antibiotic resistance gene has to be inserted.

CRISPR genes are generally 21-48 bp long and insertion of new spaces shows mutation and in turn resistance against phages. The mutation could be caused by single nucleotide mutation also.
