Question

I) Which two sterile techniques allow you to separate cells in a culture into individual colonies? Explain why these techniques can accomplish this separation. (4 pts) 2) In the inoculation experiment, do you expect growth in the control tube? Explain your answer. In your answer, include what it would mean if there were growth in the control tube. (2 pts) 3) Consider the following results: Strain Minimal medium no growth growth no growth Minimal medium + adenine no growth growth growth (a) Can strain 2 make its own adenine? Explain your answer. (2 pts) (b) Can strain 3 make its own adenine? Explain your answer (2 pts) (c) What situations might explain the results obtained with strain 1? Provide two different hypotheses. Consider both possible technical problems and possible nutritional requirements of the strain in formulating your hypotheses. Propose a nutritional test/experiment that could differentiate between the two possibilities. (4 pts)

Explanation / Answer

3.a) Strain 2 can make its own adenine. When we compare it with minimal medium & minimal medium+adenine, strain 2 can grow in minimal medium. Thus it can make its own adenine.

b) Strain 3 can't make its own adenine. When we compare it with minimal medium & minimal medium+adenine, strain 3 can't grow in minimal medium. Thus it can't make its own adenine.

c) Hypotheses 1 (Nutrient requirement): Strain 1 requires more nutrients to grow. We can use different media for this purpose.

Hypotheses 2 (Technical error): We may have done error while making media or the inoculation isn't properly done.

To differentiate these two possibilities we can repeat the experiments to determine if it is due to technical error or not. If the same result persists then it's not due to technical error & we can try different media. If this time strain 1 grow then our hypotheses 1 is correct.
