The pH of the environment can affect bacterial growth, depending on the type of bacteria it may require a very acidic area to thrive in, meaning it can only survive in low pH environments. Some species of bacteria have no chance of surviving in an acidic pH due to denaturation. You can test the acidity with a litmus paper, by dipping it in the bacterial culture and analyzing the color change of the paper.  Temperature can also effect bacterial growth, of course test this by heating and cooling the culture, and then seeing if the culture grows or dies.
