Question

Answer the following according to the graph below:

a. Is the graph for all four samples similar over the full 3 minutes? If not, why do you think this is?

b. The rate of the reaction is determined by the initial part of the reaction where it is linear i.e. the slope of the line. Is the rate of oxidation of guaiacol the same for all of the peroxidase enzymes? If not, what do you think the reason for this could be – there may be several?

Peroxidase Assay of 4 Plant Tissues 4.5 3.5 Potato Sweet Potato Radish Brussel Sprouts 2.5 ^ 1.5 0.5 0 40 80 100 120 140 160 180 200 20 60 Time (s)

Explanation / Answer

a. The graph reveals that among the four plant tissues taken, peroxidase is maximum in brussel sprouts and minimun in potato. The graph is revealing that the amount of peroxidase concentration is increasing with time. But in brussel sprouts its increase is maximum.

b... Guaicol reacts with H2O2 to form tetraguaicol and 8H2O. Absorbance is checked at the concentration of tetraguaiacol. Greater the concentration, greater will be the absorbance.

Rate of oxidation = [guaicol]4 [H2O2]2

The equation itself states that the rate is not same for all four tissues.

Reason:- Less hydrogen peroxidase production will give less rate of oxidation, even though same amount of guaicol is added in all the four reactions.
