1). DNA polymerase I is having 5’ to 3’ exonuclease activity, which restricts its use in many biological applications. Klenow fragment is a protein fragment derived by the enzymatic cleavage of DNA polymerase I of E.coli. This fragments retains the polymerase activity in 5’ to 3’ direction and also 3’ to 5’ exonuclease activity, which facilitates proofreading and also removal of precoding nucleotides. Klenow fragment does not contain the 5’ to 3’ exonuclease activity.
