Question

Can someone please confirm my answers to the following questions. Thanks in advance.

1) If tissue sections are of poor quality due to inadequate fixation, the problem may be corrected by refixing the tissue. True: False___

Unfixed or poorly-fixed tissue will be damaged by tissue processing. However, gentle reprocessing that includes some additional fixation is probably the best compromise. So refixing the tissue, say, in paraffin, would correct the problem?

2) The preservation of biological reactivity by an aldehyde fixative is inversely proportional to the number of cross links formed and the rate at which they are formed. True___ False:

I believe the more cross-links are formed at a higher rate would optimize preservation of specimen; so, The preservation of biological reactivity by an aldehyde fixative is NOT inversely proportional to the number of cross links formed and the rate at which they are formed so the answer is FALSE, correct?

Explanation / Answer

Answer

1) your first answer is right as poorly fixed tissue can be re-fixed in order to improve the tissue section quality. So above statement is right

2) In my opinion, your second answer is not appropriate because the degree of crosslinking of protein is inversely proportional to the biological reactivity. Biological reactivity of specimen depends on the structural integrity of the protein epitope present in the specimen. If a number of crosslinking will be more then the probability of crosslinking of epitope will be high which may hamper the structure of epitope that eventually may decrease the bioreactivity of the sample. So there is a need of optimum crosslinking in order to fix the tissue.

So statement 2 is true.

Hope this explanation will help to clear your confusion.
