If a bacteria with an Optical Density (OD) of 0.25 prior to plating formed 100 colonies when 0.20 ml of the culture was added to a culture plate, how many colonies would form if 0.10 ml of the same culture was added to a culture plate?

Explanation / Answer

0.2 ml of the culture contained 100 colonies. Since the same culture is used, 0.1 ml would obviously contain half the number of colonies of 0.2 ml culture, i.e, 50 colonies.

Whatever the optical density, we used the same culture. Therefore we can calculate directly.
