Question

Please answer the following question fully and completely!

3, You are trying to isolate an enzyme that catalyzes lyse cells conversion of A to B, and you have a very sensitive assay to for this enzyme. After lysing the cells, you find that the activity is associated with the membrane fraction, nut with the soluble (cytosolic) fraction, which you discard. You no activity activity then follow the rest of the procedure as shown. Explain pH from 7 to 6 why activity sudden returned when those two fractions were combined. soluble trash no activity no activity soluble no activity no activity activity combine fractions

Explanation / Answer

Above given information, we need to isolate enzyme from bacterial cell and enzyme activity associated with membrane of bacterial cell, after breaking cell we get two fraction 1. Membrane fraction or 2. Soluble cytosolic fraction. After lysis we know our enzyme embedded in membrane, and enzyme is very sensitive to pH, Temp. and another cytosolic fraction have no enzyme means no activity but it matter because its give favorable environment for enzyme to give high activity so that enzyme can convert A to B product. Cytosolic faction maintain pH inside the cell,

Normally membrane enzyme show activity if we change in pH than it loss its activity, but if we mix both fraction , give full activity of enzyme and convert substrate into product
