Question
Please give an explaination for the answer as well.
1. Following dilution of a concentrated solution, the absorbance is determined to be 1.635 on an instrument similar to the Spec20 used in this lab. What, if any, changes should be made to this experiment?
e. There is nothing wrong with this analysis and the 1,635 should be recorded
a. The sample should be diluted at least two fold b. The sample should be concentrated 2 fold c. The sample should be diluted 100 fold d. The sample should be put into a different cuvettee. There is nothing wrong with this analysis and the 1,635 should be recorded
Explanation / Answer
According to the equation , A=log10 Io/I
Abs value 1.635 means only 2.3% transmittance in light, 97.7% of light is absorped. Measurement with very high absorbance value is not recommended, since it increases the chance of error. Your sample allows only very small amount of light to pass which may incease error in measurement.
In my opinion you should try to keep the measurement value between 0.2-0.9. But some instruments can measure up to 2.0 unit absorbance. In that case you should check if this value is linear in this area.
If not, you should use more dilute solution. Try 100 times dilution. Check if that lies between the preferable range of 0.2-0.9 .
