Question

Please explain in detail how to get the answers below. They are all correct. Thank you!

15. (12 Points) Pollen (male gametes) from a plant with red flowers was used to pollinate ovules (female gametes) of a plant with white flowers. All F1 plants had red flowers. When F1 plants were backcrossed to the parental white flower plant generate F2 generation, you observed 1 red: 1 white ratio. In addition, an RFLP (restriction fragment length polymorphism) pattern for the parents, F1, and ten F2 individuals with white flowers is shown below arents Ten F2 plants with white flowers red white F1 9 kb a. Is this RFLP linked to the gene that determines flower color? Briefly explain your answer. (4 points) Yes, because RFLP ratio is not 1:1 b. If they are linked, what is the map distance between them? If they are not linked, check the box below and provide a brief explanation. Show your work and circle your answer. (4 points) 2/10-20% c. If there are any abnormalities in the RFLP result, please briefly explain. (4 points) -The last F2 plant didn't receive a chromosome from the white flower parent. -Non-disjunction happened in the white flower plant (during either meiosis I or meiosis II)

Explanation / Answer

a.. yes, it is linked with the gene. Because the gene for red color has the restriction site which cleaves the DNA and that restriction site is not present on white colored gene, for conforming the result that the gene is cleaved is our gene of interest, the F1 heterozygous has all three bands which means two red colored cleaved band and one intact not digested white colored gene band observed. No other band is observed this represents that the digested gene is gene for red color only.

b.. here as you answered, the recombination frequency is 20% . recombination frequency more than 50% indicates that both gene are not closely related or war away from each other or may be on different chromosome. 20% is the RF of our genes which indicates that genes are closely related. Distance between both gene is 20cM.

C. yes, 3rd last lane has three bands as like F1 generation. Hence it will be red (as all F1 are red) additionally the last one has only digested gene for red color and white color gene is absent. This is due to the non disjunction which leads to polyploidy in plants. Polyploidy results in chimera, some cells have that chromosome for gene and some do not have. cells in poly ploidy have abnormal number of chromosome, Due to the polyploidy we had observed these type of band pattern in these 2 cases.
